The CSC theory has been widely recognized due to thecontinual emergence of new evidence. At present, the origin of theCSCs remains unclear, but emerging evidence suggests that CSCs playan important role in the occurrence and development of malignanttumors. Tumor stem cells have also been considered as the majorcause of drug resistance to chemotherapy and radiotherapy failure(,–).Surface molecular markers can be used for flow cytometry screeningof a unique subset in a heterogeneous population of cells such ascancer cells. CD133 is expressed in a variety of tumors as arecognized marker for tumor stem cells such as brain tumor(,), breast cancer () prostate cancer (), colon cancer (), pancreatic cancer (), hepatocellular () and renal carcinoma (). However, CD133 remains highlycontroversial as a tumor stem cell marker (–). Aprevious study showed that both CD133 andCD133 subpopulations of A549 and H446 cells containcancer-initiating cells (),which indicated that CD133 may not be an ideal marker for lungCSCs. Therefore, as the identification of lung CSCs is fundamentaland vital for further research, for example, the mechanism of lungCSCs involved in the drug resistance to chemotherapy andradiotherapy, more specific and accurate markers for identifyinglung CSCs are urgently required.

This study will determine if there’s a significant difference between LED and LCD monitors.

Brainstorm the topic
What are your concerns? What concerns are facing your field of study?
For example, you are in construction and you are concerned with the slow recovery in New Orleans after Katrina. You begin the thought process like this:

11. What is a proposal anyway? A good proposal should consist of thefirst three chapters of the dissertation. It should begin with a statementof the problem/background information (typically Chapter I of the dissertation),then move on to a review of the literature (Chapter 2), and conclude witha defining of the research methodology (Chapter 3). Of course, it shouldbe written in a future tense since it is a proposal. To turn a good proposalinto the first three chapters of the dissertation consists of changingthe tense from future tense to past tense (from "This is what I wouldlike to do" to "This is what I did") and making any changesbased on the way you actually carried out the research when compared tohow you proposed to do it. Often the intentions we state in our proposalturn out different in reality and we then have to make appropriate editorialchanges to move it from proposal to dissertation.

17. The major myth in writing a dissertation is that you start writingat Chapter One and then finish your writing at Chapter Five. This is seldomthe case. The most productive approach in writing the dissertation is tobegin writing those parts of the dissertation that you aremost comfortable with. Then move about in your writing by completing varioussections as you think of them. At some point you will be able to spreadout in front of you all of the sections that you have written. You willbe able to sequence them in the best order and then see what is missingand should be added to the dissertation. This way seems to make sense andbuilds on those aspects of your study that are of most interest to you at any particulartime. Go with what interests you, start your writing there, and then keep building!

CD90 (Thy-1) is a 25–37 kDaglycosylphosphatidylinositol (GPI)-anchored glycoprotein expressedmainly in leukocytes and is involved in cell-cell and cell-matrixinteractions (). CD90 expressionwas identified in murine breast CSCs (), primary high-grade glioma CSCs(), and in liver malignancy(). Our previous study () initially demonstrated that the A549tumor sphere cells had a stronger capacity for proliferation andself-renewal, and expressed higher levels of stem cell markers Sox2and Oct4 than A549 adherent cells. In the present study, we carriedout flow cytometry analysis and other experiments to explorewhether CD90 could be a marker for identification of lung CSCs.

Human lung cancer cell lines A549 and H446 (ShanghaiInstitute of Cell Biology, China) were used in this study. Thecells were cultured in RPMI-1640 medium (Gibco, USA) supplementedwith 10% fetal bovine serum (FBS) (Gibco). The tumor spheres werecultured in serum-free conditioned medium that contained DMEM/F12medium (Gibco), 20 μl/ml B27 supplement (Gibco), 20 ng/ml basicfibroblast growth factor (FGF) and 20 ng/ml epidermal growth factor(EGF) (both from PeproTech, USA). All cells were incubated at 37°Cwith 5% CO and 100% humidity. The third-generationsphere cells were used for further experiments.
