The data just presented demonstrate that pyoverdineB10, as the end product of its biosynthesis pathway, is a key participant in the control circuit regulating its own production in Pseudomonas strain B10. In P. aeruginosa PAO1, however, extracellular pyoverdinePAO1 has no apparent role in the positive regulation of the homologous pvdA gene see Fig. for a comparison between the two systems). The lack of pvdA response to exogenous pyoverdine is consistent with the observation that pvd mutants blocked in late steps of pyoverdinePAO1 biogenesis produce wild-type levels of hydroxamate nitrogen in iron-poor medium (). This implies that PvdA is equally expressed in the pyoverdinePAO1-proficient and -deficient backgrounds, and immunoblot analysis of PvdA expression in the wild-type and pvd mutants corroborates this conclusion.

Strains of P. aeruginosa secrete one of three pyoverdines (types I to III) (). Genes and enzymes required for pyoverdine synthesis in strain PAO, which secretes type I pyoverdine, have been characterized experimentally (, , , , , , , , , ). This has revealed a biosynthetic pathway in which a pyoverdine precursor is assembled by nonribosomal peptide synthetases (NRPSs), with other enzymes providing the unusual amino acid substrates for the NRPSs and modifying the precursor peptide to yield the mature pyoverdine. Genomic analyses imply that the pathway of synthesis is similar in outline in other Pseudomonas species (), and this is supported by experimental evidence (, , ). Pyoverdine synthesis has not been studied experimentally in strains of P. aeruginosa other than PAO1. However, recent genomic analysis shows that strains that make different pyoverdines share many pyoverdine synthesis genes with strain PAO, but they contain additional genes that are not present in strain PAO and are proposed to be required for pyoverdine synthesis ().

We investigated the regulation of the psbA and pvdA pyoverdine biosynthesis genes, which encode the -ornithine N5-oxygenase homologues in Pseudomonas strain B10 and Pseudomonas aeruginosa PAO1, respectively.
