Irini Pateraki tries to elucidate the natural biosynthetic pathway of the medicinal drug forskolin in the plant Coleus forskohlii. She recently was author on a featured in the Journal Plant Physiology showing the first results of her project along with in planta localization studies for forskolin.

14/03/2017 · The entire biosynthetic pathway for the diterpenoid forskolin in Coleus forskohlii has been reconstituted in yeast.

The next steps of our research are firstly to express forskolin biosynthetic pathway in heterologous hosts for sustainable production of forskolin, but also to study the transcriptional regulation of the different diterpenoids pathways in Coleus plants.

Why did you choose to work with this? / with synthetic biology?
I started studying classical plant biochemistry during my bachelor and master and expanded more into plant genetics during my PhD by working on the genetic model plant Lotus japonicus. In lotus I primarily studied UDP-glucosyltransferases involved in the biosynthesis of hydroxynitrile glucosides which includes cyanogenic glucosides. This lead me back to doing biochemistry as my knowledge about UGTs made me collaborate with , who studied metabolon formation in the dhurrin pathway of Sorghum bicolor.

The basic science that we do on the model compounds cyanogenic glucosides can be transferred to the biosynthesis and production of valuable compounds, which can be used within the food, feed and medicinal industry and thereby be of value to the society.

The expression of 17-ketosteroid reductases in the adrenal has also not been investigated in depth, largely due to neither 17-hydroxy-C19-steroid nor estradiol secretion is commonly associated with the human adrenal cortex. Therefore in the present study we describe our initial studies to evaluate the putative pathway involving both aromatase and 17-ketosteroid reductase in the biosynthesis of the active estrogen, estradiol, in H295 cells. We have also compared the expression pattern of steroidogenic genes observed in H295 cells to the patterns observed in two distinct adrenocortical tumors. The products of the first tumor were believed to promote feminization (gynecomastia, loss of libido) in an adult male since these features were resolved after a successful adrenalectomy. The second tumor was considered to be an aldosterone-producing adrenal adenoma based on the clinical and biochemical history, and the post-operative remission of hypertension and hypokalemia.
